Not to mention how I think the night before Mukuro's body was discovered and the 5th trial, Makoto was inexplicably sick. It could've been food poisoning since it'd be strange for him to get a viral illness in a completely sealed off school, but what if Junko somehow poisoned him in some way to bring him to his lowest to make his attempted murder easier for her to carry out?

Whatever it was, definitely was used to make him physically and mentally weaker.

He was ill and he was restless all night. Meaning he also got no sleep.
And is probably why he took such a long nap in the rubbish heap, he's had a long day.
If Makoto is sick and tired he's more likely to make mistakes, miss things in the trial and also be an easier target.
And these all could be noted as symptoms of posion.
Either through his food or simply just through the vents in his bed room.
