MHA 2.2 - Roaring Sports Festival - part 3
Tumblr media
Pffft. This guy. I am a socially anxious mess but I would 100% party with this man. It would be impossible to feel self-conscious next to him.
Tumblr media
And while we are on the topic of anxiety, being stared at by so many people is nauseating and terrible. Blegh. This is a lot of pressure.
Tumblr media
Haha, the sad/funny part is they are right. The basic background character designs compared to Purple People Eater's obviously 'important' design is hilarious to me. Like they are all beige and boring and upfront is this beautiful purple monstrosity. Guess which character you need to pay attention to?
Tumblr media
Speaking of monstrosities, what the hell? This show has been a bit up and down with female representation so far, and to keep it short, this lady is getting filed in the down category. The sexy high school teacher fetish, I mean, really?
Tumblr media
Thank you for asking. The answer is NO.
I wonder if Birb Boy is also blushing, but we can not see is due to his black plumage. Just bird life things.
Tumblr media
Ohohoho, this should be good, and by "good" I mean a garbage fire.
Tumblr media
BOO LOUDER! You know what bothers me about our grumpy Pomeranian, more than his general abrasiveness, is how self-centered he can be. It is always "me" and "I" with this kid.
Tumblr media
It is not wrong to be confident or to be ambitious. It IS wrong, however, to put others down to make yourself feel superior.
I just hope that Bakugo's character develops from here and he isn't just like this forever. That would be unbearable to keep watching. Even if him being pissy is funny at times.
Tumblr media
Deku is trying to tell me that Bakugo has grown somewhat since he is being serious here. But I'm gonna need a little more than that to be convinced.
Not going to lie, I picked this screenshot mostly for Iida in the background. Look at them hands.
Tumblr media
HA! The way Izuku looks at the camera like he is in The Office. He is so done with Bakugo's bs.
Still, Iida is a natural pop & lock dancer. He missed his true calling in life.

@ people who redraw bnha characters as brown/black and claiming its for poc diversity
please remember that these characters are asian
being asian means they are people of colour
and please just say that you redrawing them as brown/black isn't for poc diversity but for watching the world burn and hating on an author

I’m worried about them
Tumblr media
Summary:  You’ve been a bit off since you got out of the hospital.
Pairing(s)/Character(s): Dekusquad/Reader
Word Count:  565
Warning(s): Gender Neutral reader (uses they/them pronouns), mentions of a hospital stay, anxiety, panic attack
A/N:  I missed writing and this gnawed at my brain for a couple of days before I wrote it.  Yes, this is my first time writing for My Hero Academia, so the characters might feel a bit off.
I know it’s a bit personal but this is how I felt after I got out of the hospital.
Beta’d by the lovely @iflostreturntosteverogers.  Dividers by @firefly-graphics
"How is Y/N actually doing?"  Izuku asked Shoto as he placed the plastic bags full of food on the kitchen table.  He was concerned with how you had been acting since you got out of the hospital.  You weren’t acting like yourself.  You avoided your room on the second floor like the plague and every night, you slept in one of their beds.
"They had a rough night sleeping.  Kept waking up every couple of hours, and I'm pretty sure that last time was because they had a nightmare but they wouldn't talk to me about it.  Just curled right back up into my side and stayed there until my alarm went off."  Shoto frowned as he brushed a few stray hairs back from your forehead.
"I'm really worried about them.  Do you think we should take tomorrow off, and spend the day with them?"
"I think they'd like that.  I know they haven't said much, but I think being alone during the day is making their anxiety flare up."  Izuku blinked in surprise when Ochako answered instead of him.  That's when it hit him, he still had groceries to help carry in.
He made his way towards the door when Iida and Tsu stepped into their shared house with both arms full of grocery bags.
"Sorry guys, I got distracted."  They waved their hands in tandem as if to say it was fine.
"Next time, you'll be responsible for bringing everything in."
"But for now, kero, we need to put all of this up and start on dinner."
"That'll be a fantastic way to wake them up!"  Ochaco clapped her hands together as she grabbed a couple of the bags dangling from Tsu's arms and started putting stuff away.
You rolled off of the couch and landed on your side.  A small 'oof' was heard but no one rushed into the room.  You sighed and rolled onto your back.  Where was everyone?
You stared up at the ceiling and a thought crossed your mind.  You fell asleep in Shoto's lap, right?  So where the hell did he go?  Why would he leave you like that?  Was he tired of you already?  You were a tad clingy ever since you got out of the hospital, you knew that.
"Whoa, whoa I can hear your mind racing from here, Y/N.  Shoto is making up a plate of food for you, he didn't leave you.  None of us are leaving you."
"Are you sure?"  You slowly sat up and met Izuku's soft gaze.  He took a seat beside you and gently tugged you so that you could sit between his legs.
"We love you too much to let you go just like that."  He pressed a kiss to your temple and you took a deep breath in.  A few seconds passed and you released it.  "There we go, just keep doing that okay?"
You timed your breathing with his heartbeat and it was like time was moving slower until you felt yourself calm down.  “Zuku?”
“Yeah?”  The words left you as soon as you called out his name so you did the only thing you could think of.  You pressed your lips against his cheek and was planning on thanking him, but your stomach growling interrupted that.
“Okay let’s eat dinner and watch a movie afterwards, does that sound like a plan?”
“That sounds perfect.”

It smells so good
Tumblr media
Summary:  While sorting out your laundry, you find that some of your clothes smell different.
Square Filled: Scent Marking (ABO)
Pairing(s)/Character(s): Alpha! Dean/Omega! Reader (eventually),  Rin (OFC)/Ash
Word Count:  656
Warning(s)/Rating: Teen+ | Secret keeping from reader, reader has an admirer.
A/N:  This was written for @spnabobingo.  The photos I used for my aesthetic came from Pinterest.
Beta’d by the lovely @iflostreturntosteverogers
Part 1 of 2
You eyed the overflowing laundry basket in the corner and sighed. You needed to do your laundry. Hell, you needed to do anything that wasn’t laying in your bed and scrolling through TikTok. You checked your phone battery before locking it and tucking it away in your pocket.
You hauled yourself up and out of the bed.  As you shuffled across the room, you threw the clothes that didn’t reach your basket onto your bed.  You snaked your hand around the plastic handle of the basket and pulled it over to your bed.
You dumped the basket out and started sorting them.  Your familiar scent of coconut, aloe, and mint was all you were smelling until you got to your favorite shirt.
Bringing it up to your nose, you sniffed at it and your nose scrunched up at the vaguely familiar scent.
“What are you doing?”  You peered over your shoulder to see your roommate standing in the doorway with her phone in one hand and a gift bag in the other.
“I know this might sound like a weird request, but will you smell this?”  You held up the shirt and waited for Rin to approach.
“For the record,” Rin answered with a chuckle as she rested her chin on your shoulder and got a good whiff of the shirt in question.  “This is normal compared to the other stuff you’ve had me do.”
“Rin….”
“When were you around an Alpha?”  She asked in all honesty and you shrugged.  You had no idea.
"I haven't-"  You paused and shook your head.  "The last Alpha I was around was your boyfriend."
Rin snorted.  "Dean is not my boyfriend."
"He's not?"  Your inner omega was practically leaping for joy with this information.
"Dean's hot but he's not my type."
"I forgot that you like them average sized, soft and with a kickass mullet"  You snorted out a laugh and tossed the shirt in your hands onto the bed with your other dirty laundry.
"Damn right I do."  Rin grinned as she dropped the gift bag she was holding into your lap.  The same scent entered your nostrils and you raised an eyebrow at the gift.
"It's the same, I'm sure of it."  You muttered to yourself.  "It smells so good."
"Well open it!"  You peered into the bottom of the bag, grateful that whoever sent you this gift hadn't loaded it down with tissue paper.
The first thing you pulled out was a simple white card with a picture of a sunflower on it.  You opened the small card up and read the simple message inside.
I hope the next time we meet you'll be wearing this.  Maybe by then, I'll have the courage to show myself. - D.W.
You made a note to try and figure out who D.W. was later.  For now, you reached into the bag and pulled out what looked like a black shirt until you unfolded it.  This was the dress I was looking at last week…

I love everyone in Boyfriend Dungeon except Eric, Masamune, and Issac's father.
Solid game.
Do I regret obsessively playing it? No.
Am I sad that it took me 2/3 days to beat it? Yes.
Am I sad that it said there was 9 weapons you could date, and only 7 are in the game? Also yes.
The game is fantastic. The music is great and the characters ugh. I love them all. Just don't make me pick a favorite bc I can honestly say I'm gay for them all 😂😂😂

Have a good nap, Dean
Tumblr media
Summary: Dean shows that he cares for you in a different way.
Pairing(s)/Character(s): Dean Winchester/Reader
Word Count:  183
Warning(s)/Rating: General | None really.
A/N:  Have a drabble that I wrote out at 7 this morning.  No beta here lol so all mistakes are my own.
The photos I used for my aesthetic came from Pinterest.
Dean Winchester does not show his love in a normal way.  Or at least that's what you think.  When you stop and think, it's the little things that he does throughout the day that show you how much he cares.  The two of you are teetering into "I love you" territory, but you're both stubborn and neither of you will make the first move.
That's when an idea hits you.  You spend a lot of time occupying yourself while the boys are on hunts, so you march down the hall to the Dean cave and linger in the doorway.  "Hey Dean?"
You wait for a minute but there's no response, so you step into the room and move closer.  You call out his name again, and once again, he doesn't answer.  You sigh softly and move closer to the recliner and see that Dean is stretched out, fast asleep.
Your idea could wait.  Letting Dean rest was higher on your list, so you leaned down and pressed a kiss against his forehead.  "Have a good nap Dean."
"It'd be better with you here."

Tamaki HC’s | MHA
Tumblr media
Request: Can i ask for an headcanon for how Tamaki Amajiki would get close to his crush who works with him and Fatgum as well?
Word Count: 774 words
Page Count: 2.2 pages
Authors Notes: a reupload from my old blog, hope you all still like it!
🦑  Tamaki Amajiki / Suneater  🐚
🦑 You’re a year older than him at this point, moving from America to Japan to take care of your grandparents who had lived there for the past 30 years, taking a job at Fatgums hero agency as well. Your quirk was shape-shifting, but the official name for it was Transform, the reason why you don’t know :/
🐚 You’re scheduled for a night patrol on a Thursday night, and when you’re getting ready you get a call from Fatgum to meet up at one of his favorite places to snack at, saying he needs a favor.
🦑 You’re down, you’re chill. You meet up.
🐚 You walk down the street and see him, and another hero by his side, who is shaking lightly. You felt kinda bad and wondered if he was okay.
🦑 “Hey Gummy!” You smiled, waving as you were a few feet away from him, causing both of them to turn around.
🐚 "Hey Honey!“ He greeted back, before looking to Tamaki and oh boi
 🦑He’s star struck
🐚 His heart went doki doki or sum shii ( lmao what am i doing )
🦑 He’s staring at you, your costume being just some black leggings and a tank top, with a leather jacket and he just can’t stop
🐚 You tried introducing yourself to him but he just can’t sputter out anything
🦑 Fatgum just chuckles before explaining what he needed
🐚 It was a school night for Tamaki and he didn’t want the kid to overwork himself, so he just wanted you to help him out, and not let Tamaki stress himself out too much.
🦑 Okay, no problem, and maybe you can help this kid calm down you thought
🐚 The ENTIRE NIGHT he struggled to talk to you
🦑 Nothing eventful happened, other than a small girl getting lost from her father
🐚You helped calm the poor girl down, and carried her around while Tamaki alerted other heroes on patrol that he found a girl looking for her father
🦑 He watched you interact with her for a while, admiring how good you were with the kid, even mocking your own American accent when she asked why you spoke a bit funny
🐚 His heart was TOUCHED
🦑 You ended up buying some snacks for the girl and Tamaki
🐚 TOUCHED 2.0
🦑 You both ended up finding her father, who thanked you both so much for taking good care of his daughter
🐚 You smiled and only said it was the right thing to do
🦑 The night ended around 12, and you walked Tamaki back home, and had a small conversation
🐚 You noticed how he had communicating issues because he is anxious, so you took the lead
🦑 You spoke about your family and past, trying to get him more comfortable with you, and he would pitch in and you would just LISTEN
🐚 He appreciated it so much
🦑 You gave him a pat on the shoulder and ruffled his hair before he went to his house, face REDDER THAN KIRISHIMAS HAIR
🐚 Once his bedroom door closes he jumps into bed and screams into the pillow, he’s just OOF
🦑 You ended up doing this more and more, patrolling with him on school nights, exchanging numbers and talking more
🐚 You were really patient with him and he just loves that so much
🦑 Silence with you was never awkward, it was comforting
🐚 He would start to text you more often, little good mornings and good nights
🦑 Send you little funny and cute things, from cat gifs to whole ass cryptic memes about weird shit
🐚Usually about anxiety, but he tries, and you know this and you love it
🦑 He would try and buy you some snacks too, because tbh food is the only thing that makes everyone happy
🐚 He would also lay his head on you a lot, it means so much to him that you’d let him, he trusts you
🦑 Fatgum cackles manically in the corner as he watches young love bloom
🐚 He takes full responsibility for this lmao
🦑 Tamaki would do small little things to show he is trusting you and is starting to fall for you
🐚 Bringing you food, holding pinkies, and sometimes when things are getting rough he tries to cuddle you if it helps
🦑 Its just too pure
🐚 oml
🦑 We all need a Tamaki tbh

Totally Clean Anthem House? Not Without Air Duct Cleaning
Totally Clean Anthem House? Not Without Air Duct Cleaning
Air duct cleaning in your Anthem home is an investment in the air quality and the protection of the health of its occupants. Regularly maintaining your HVAC system and changing the filter helps. However, there is still a buildup of dust and airborne pollutants that gets into your ductwork and professional cleaning is the best way to remove it. Here are five good reasons to have it done regularly.

People preferred hanging over other ways to end lives in 2019-20 - NCRB
People preferred hanging over other ways to end lives in 2019-20 – NCRB
Srinagar, Nov 12: Most of the people who had committed suicide during the fiscal 2019-20 in Jammu and Kashmir have adopted hanging as a tool to end their lives. According to the official figures of National Crime Records Bureau for Ministry of Home Affairs (MHA), out of the total percentage of suicides committed across India, Jammu and Kashmir has a 2.2 percent with most of the victims using…

MHA 2.2 - Roaring Sports Festival - part 4
Tumblr media
They are going to act like the sport commentators? This is perfect.
Tumblr media
And off we go! We already have some clear leads. Todoroki is just making the whole race his personal ice skating rink from the get go.
Tumblr media
Haha, I am kind of mad I did not think of the half and half nickname myself. Rage on, pissy teen, rage on. Bakugo: 1. Me: 0.
Tumblr media
The show seems to be setting this purple muppet up to be a suspicious antagonist type, but I can not look at this and see anything but KING behavior.
My only question is how does his quirk make this happen? I'm guessing he is controlling them. Either that or his quirk is to compel people with an insatiable urge to give him piggyback rides. In which case, best. quirk. ever.
Tumblr media
Don't be underestimating. Frosty has a cool quirk but he is not the only one. I actually think the girl who can pull murder weapons out of her body has the quirk with the most potential.
Tumblr media
As opposed to a Disco Inferno! So, the storytellers are using the big scary robots and have Frosty take them out easily to build up how strong Todoroki is. I wonder why the show is focusing on him and even starting to pit him against Deku. Hmmm.
Tumblr media
Old man? Oh nooo, don't tell me. Incoming Daddy issues? Amirite?
Besides that, Zuko's long lost twin, is looking hella cool (no pun intended) in this shot. Just look at him with his cascading snowflake sparkles.
Tumblr media
Strike a pose, vogue, vogue, vogue, vogue...
I love how determined he is, but I wish I knew more about why he is determined and for what goal.
Tumblr media
Welp. Mama is not holding her shit together. It is a good thing she never saw footage of the USJ attack if she is sobbing over seeing her kid participate in a sports race.
Tumblr media
I love Deku's conflicted expression. He is excited, nervous, and determined all in one, and the artist managed to convey all of that in his face here. I just think that is neat.
I wonder who will win the race?

Sage Therapeutics and Biogen Announce Positive Pivotal Phase 3 Results for Zuranolone, an Investigational Two-Week, Once-Daily Therapeutic Being Evaluated for Major Depressive Disorder
Tumblr media
CAMBRIDGE, Mass.–(BUSINESS WIRE)–Sage Therapeutics, Inc. (Nasdaq: SAGE) and Biogen Inc. (Nasdaq: BIIB) today announced that the WATERFALL Study in patients with MDD met its primary endpoint with zuranolone (SAGE-217/BIIB125) 50 mg showing statistically significant improvement in depressive symptoms compared with placebo at Day 15 as assessed by the 17-item Hamilton Rating Scale for Depression (HAMD-17) total score. LS means (SE) change from baseline in HAMD-17 total score at Day 15 for patients who received zuranolone 50 mg was -14.1 (0.51) compared with -12.3 (0.50) for patients who received placebo (LS mean difference -1.7 points; p=0.0141).
Monoamine-based antidepressants have been the standard of care for chronic treatment of MDD for the past 60 years. They are treatments administered daily, which require sufficient exposure and continuous use to maintain effect. Zuranolone is a two-week, once-daily oral drug under investigation for the treatment of MDD. It is a molecule that is designed to potentially provide a rapid-acting, sustainable treatment option, and could represent a breakthrough in the current management of depression.
The WATERFALL Study was a pivotal, Phase 3, double-blind, randomized, placebo-controlled study evaluating the efficacy and safety of zuranolone 50 mg in adults 18 to 64 years with MDD (N=543). The WATERFALL Study enrolled patients who had MDD with a HAMD-17 total score ≥24 at screening and Day 1 prior to dosing.
“Sage’s expertise in the modulation of the GABA receptor pathway in the brain, coupled with insights on the treatment wants and needs of clinicians and patients, has resulted in our targeting a unique benefit/risk profile with the development of zuranolone supported to date by the data generated in the WATERFALL Study and the broader Landscape and NEST programs,” said Barry Greene, Chief Executive Officer at Sage Therapeutics. “We dared to imagine a different future for the treatment of MDD where patients have the potential to experience a rapid response that is well-tolerated and that may enable them to stay better with long periods free from depression symptoms, and free from daily chronic treatments and related side effects. In doing so, we aspire to help eliminate stigma associated with brain health disorders so that we can move beyond brain health awareness to brain health action.”
“Together with our collaboration partners at Sage, we are proud to announce highly encouraging results from the Phase 3 WATERFALL Study of zuranolone in major depressive disorder. These results represent hope and positive progress for the more than 250 million patients worldwide who are estimated to live with depression,” said Alfred Sandrock, Jr., M.D., Ph.D., Head of Research and Development at Biogen. “Major depressive disorder is a common co-morbidity of many diseases represented in Biogen’s neuroscience portfolio. We believe zuranolone has the potential to offer a unique, first-in-class therapeutic for depression with a distinct benefit-risk profile to people living with this common but serious mental health condition.”
Zuranolone was generally well-tolerated in the WATERFALL Study and demonstrated a safety profile consistent with previous clinical studies. The rate of treatment emergent adverse events (TEAEs) in the zuranolone group was 60.1% (161/268) vs the placebo group at 44.6% (120/269). The majority of the TEAEs were mild to moderate. The most common TEAEs that were ≥ 5% in patients treated with zuranolone (rates vs placebo) included somnolence 15.3% (vs 3.0%), dizziness 13.8% (vs 2.2%), headache 10.8% (vs 7.8%), and sedation 7.5% (vs 0.4%); these events predominantly occurred during the 14-day treatment period. Throughout the study, a total of two patients each (0.7%) reported serious adverse events (SAEs) in the zuranolone and placebo groups; no death occurred in the study. The percent of patients reporting TEAEs leading to drug discontinuation was 3.4% (9/268) and 1.5% (4/269), in the zuranolone and placebo groups, respectively. No signal for withdrawal symptoms as assessed by the 20-item Physician Withdrawal Checklist (PWC-20), or for increased suicidal ideation or behavior as per the Columbia-Suicide Severity Rating Scale (C-SSRS) were identified.
“I’m really excited about these breakthrough data: we know MDD is episodic and zuranolone has the potential to treat episodically. The LANDSCAPE clinical studies are all helpful taken together because they provide data on both short- and long-term use of zuranolone,” said Anita H. Clayton, M.D., Chair of Psychiatry and Neurobehavioral Sciences, University of Virginia School of Medicine. “These data suggest that this treatment, if approved, has the potential to work fast with a short-course of therapy that is well-tolerated, with the effect maintained over the long-term. This will empower my patients to think differently about their depression and treatment, and to rapidly return to their life. Depression is not an identity, it’s an episodic disorder that we hope in the future to be able to treat quickly with treatments that are well-tolerated and with benefits that last.”
“MDD is a pressing mental health concern and, unlike physical health concerns where innovation is commonplace, many of the treatments for MDD were first approved more than two decades ago,” said Paul Gionfriddo, President and CEO of Mental Health America (MHA). “We welcome today’s news, and the potential for a new and innovative treatment that could change the way we treat depression.”
Zuranolone has been granted Breakthrough Therapy Designation by the U.S. Food & Drug Administration, and the Companies intend to discuss next steps with the Agency. Full data from the WATERFALL Study will be shared at future scientific forums.
Detailed Topline Results from the WATERFALL Study
The WATERFALL Study enrolled 543 patients with MDD. The patients were treated with zuranolone 50 mg or placebo once nightly for 14 days.
The primary endpoint of the study was the change from baseline in the 17-item Hamilton Rating Scale for Depression (HAM-D) total score at Day 15; the first secondary endpoint was the change from baseline in the Clinical Global Impression-Severity of Illness (CGI-S) at Day 15.
The mean (SD) baseline HAMD-17 score at entry into the study was 26.8 (2.60) in the zuranolone 50 mg treatment group (n=268) and 26.9 (2.67) in the placebo group (n=269).
90.3% of patients who received zuranolone, and 87.4% of patients who received placebo, completed the study.
Results for the primary endpoint and several topline secondary efficacy endpoints during the treatment period are outlined in the following table and all favor zuranolone:

Except for HAMD-17 at Day 15 (primary) which was statistically significant and CGI-S (first secondary endpoint) which was not significant at Day 15, all p-values in the table are nominal and not adjusted for multiple comparisons. *Pre-specified primary endpoint ^Pre-specified key secondary endpoints LS = least squares; LS mean difference = difference in LS means of change from baseline between zuranolone and placebo groups
Patients with a response at Day 15 in the zuranolone group retained on average 86.1% of their HAMD-17 improvement at Day 42 (4 weeks after dosing ended). A similar maintenance of response was also observed with the MADRS scale, where people who responded to zuranolone at Day 15 maintained 87.6% of that response at Day 42. While not statistically significant, a numerical advantage in favor of zuranolone was demonstrated at Day 42.
Safety and tolerability:
Adverse events were consistent with the safety profile of zuranolone seen to date in clinical studies.
The incidence of treatment emergent adverse events (TEAEs) in the zuranolone group was 60.1% (161/268) vs the placebo group at 44.6% (120/269).
The majority of the TEAEs were mild to moderate, with 8 (3.0%) and 3 (1.1%) being severe in the zuranolone and placebo groups respectively.
The most common TEAEs observed in ≥5% of patients in either treatment group are listed below and occurred predominantly during the 14-day treatment period. These events were non-serious, and most were mild to moderate.

Discontinuation rates of the study drug due to AEs in patients receiving zuranolone were 3.4% (9/268) compared to 1.5% (4/269) in those receiving placebo.
Throughout the study, a total number of 4 patients reported serious adverse events (SAEs), 2 (0.7%) each in the zuranolone and placebo groups.
No deaths occurred in the study.
No signal in increased suicidal ideation or behavior, as assessed by the C-SSRS, was observed throughout the study in patients receiving zuranolone 50 mg or placebo.
No signal in withdrawal effects, as assessed by the PWC-20, was observed after discontinuation of zuranolone.
No loss of consciousness, or adverse effects such as weight gain, sexual dysfunction, or euphoria were reported.
About the WATERFALL Study
The WATERFALL Study was a double-blind, placebo-controlled pivotal Phase 3 study evaluating the efficacy and safety of zuranolone in adults with major depressive disorder. In the study, 543 patients were enrolled. Patients were randomized to receive zuranolone 50 mg, or placebo, once-nightly for two weeks. The primary endpoint of the study was the change from baseline in the 17-item Hamilton Rating Scale for Depression (HAM-D) total score at Day 15. Secondary endpoints included the change from baseline in the Montgomery-Åsberg Depression Rating Scale (MADRS) and the Hamilton Anxiety Rating Scale (HAM-A) total score, among others.
About Zuranolone
Zuranolone (SAGE-217/BIIB125) is a once-daily, two-week drug in development for the treatment of major depressive disorder (MDD) and postpartum depression (PPD). Zuranolone is an investigational oral neuroactive steroid (NAS) GABA-A receptor positive allosteric modulator (PAM). The GABA system is the major inhibitory signaling pathway of the brain and central nervous system and contributes significantly to regulating brain function. Zuranolone has been granted Breakthrough Therapy Designation by the U.S. Food & Drug Administration.
Zuranolone is being evaluated as a potential rapid-acting, 2-week treatment for PPD and MDD in the NEST and LANDSCAPE clinical trial programs. The programs are designed to generate data to support a potential NDA filing as efficiently as possible. If successful, LANDSCAPE and NEST may support paths to approval with three distinct opportunities to address patient needs: PPD, acute rapid response therapy (RRT) in MDD when co-initiated with a new standard antidepressant, and as-needed treatment of MDD.
Zuranolone is being evaluated as a potential rapid-acting, 2-week treatment for PPD and MDD in the NEST and LANDSCAPE clinical trial programs. The two development programs include multiple studies examining use of zuranolone in several thousand patients with a variety of dosing, clinical endpoints, and treatment paradigms. The LANDSCAPE Program includes six studies of zuranolone in patients with MDD. Data have been reported from three studies of zuranolone 30 mg in patients with MDD (MDD-201, MOUNTAIN Study and the 30 mg cohort from the ongoing SHORELINE Study), and one study of zuranolone 50 mg in patients with MDD (WATERFALL Study). Two additional studies evaluating zuranolone 50 mg in patients with MDD are expected to read out by the end of 2021 (CORAL Study and a 50mg cohort of the SHORELINE Study).
The NEST Program includes two placebo-controlled studies of zuranolone in patients with PPD. Positive data from the ROBIN Study (zuranolone 30 mg) have been previously reported. The SKYLARK Study (zuranolone 50 mg) is anticipated to readout by the end of 2021.
About Major Depressive Disorder (MDD)
Major depressive disorder (MDD) is a common but serious mood disorder in which people experience depressive symptoms that impair their social, occupational, educational or other important functioning, such as a depressed mood or loss of interest or pleasure in daily activities, consistently for at least a two-week period. It is estimated that approximately 17 million people in the U.S. and more than 250 million people worldwide suffer from MDD each year. While antidepressants are widely used to treat MDD, large-scale studies have demonstrated the need for additional therapies with a differentiated profile.
