The subtitles don’t portray it, but he stumbles over the R sound. He was attempting to say River but got to choked up. He can’t even say it which is what makes it so powerful.

“Mama”

This is one of the most beautiful moments in the whole series in my opinion.

Jukai has cared for Hyakkimaru and given him so much love and while he has felt all this time that he has failed and only brought hurt to the boy, Hyakkimaru knows the truth, he has been able to feel it all this time and now, finally, he can put words to what Jukai is to him: A parent. His “Mama”. 

You can see how much that means to Jukai. To hear the person he has cared for as his son call him that.

It is a beautiful scene and I will always feel emotional when watching it.
