He almost immediately says he’s not, because he isn’t, but to answer the question so reflexively feels somewhat depressing. Because, without so much as a thought, he’s admitting that this really isn’t the life for him. That there’s so much more he should be doing.

Or he’s thinking too much about it, and she’s just asking whether or not he’s happy at this very moment.
