So far from the the leaks, rough translations, and the chapter summary, it is clear that:
First OFA user and Nana were testing Izuku to see his resolve regarding Shigaraki, who is 'on the path' to become the biggest evil, and if, in the case this would happen, Izuku would be ready to kill Shigaraki (especially because this would be the only way to stop him).
Izuku, however convinces that, after AM's teaching, he understood that OfA is the power to 'save people', and that deep down, Shigaraki looked like a crying for help child - which means that even if it costs him his life he will try to save Shigaraki, because this is this power's responsibility.
Deku here refers to this moment:
The 6th user name seems to be En (more on this later).
The 2nd and 3rd user of OfA have not recognized Izuku as the rightful heir to OfA, so them being against the will is a way to show their lack of approval (see here theory on their names and just general considerations on OfA users). This also means that Izuku cannot use the quirks of those who have not acknowledged him as the 9th user of OfA (2nd and 3rd in this case).
Shigaraki has been targeted by AfO because of three reasons: 1.) He is Nana's grandson (so it is a way to get back at her and OfA); 2.) Shigaraki would serve as his double, or substitute for his body (because let's remember that AfO is currently way older than it should be normally be possible, estimates around 150 years) - and the fight he had with AM just accelerated the process (this would also be a way to get back at OfA) ; 3.) So he could nurture the heatred Tenko already had towards society, and that his will would be so powerful (will to destroy) that this, in the end, would allow him to take possession of OfA.
This means that Izuku, wants to uncover the reasons behind why villains become villain, and if he can, save them (from themselves, too). And for that, he is willing to put his life on the line, especially because IT IS OfA's objective to defeat AfO, and he won't be able to do it, if he does not put the one needing saving first (especially after discovering how he is the last OfA holder).
After all, this might be one of my favourite chapters so far.
