Exposing the Wolf in Sheep’s Clothing, Cancer, with Genetic Engineering
Globally, 18.1 million people are diagnosed with cancer and there are only treatments– no cures (WCRF). The long, enduring trek to discovering the cure for cancer has been thoroughly researched but never found. Therefore, it is important to focus on more effective and comfortable treatments. CRISPR-Cas9 (Clustered Regularly Interspaced Short Palindromic Repeat- associated protein 9) genome editing is taking a new approach to treating cancer on the individual cellular level. This new technology could change cancer patients' survival rates and experiences forever.
What is CRISPR-Cas9?
CRISPR-Cas9 (with tracrRNA) is a groundbreaking, noble prize-winning technology that two women in STEM, Emmanuelle Charpentier and Jennifer Doudna, created in 2011 (Prabhune). CRISPR was discovered first as a bacterial mechanism in 1987 from E. coli and now is the name of a genome editing technique that reflects the same order of operations as it does within bacteria (Ishino). CRISPR-Cas9 has the same patterns of defense that the bacteria use to respond to foreign pathogens when fighting viruses or infections; recognized pathogens are easily remembered and defeated.

In practice, CRISPR-Cas9 is designated RNA to trace and act as "remembered pathogens" to be able to fix the problems within the DNA (Ishino). It is important to note that CRISPR-Cas9 technology is not a piece of machinery or equipment but is a protein that follows a natural process utilizing guide RNA and associated enzymes. Once a cell is inoculated with target RNA to detect the mutated gene within a different cell, it pairs with Cas9, an enzyme to aid in the cutting of the mutated DNA. CRISPR-Cas9 is then able to genetically manipulate cells by adding or removing parts of a cell's genetic material, or genome (Sarkar). The modified genome is improved, and this procedure allows for cells to be fixed and properly replicated or to help stop the duplication of bad cells.
How is Cancer the Wolf in Sheep's Clothing?
The immune system works to fight off pathogens to keep the body healthy. When the unknown invaders, antigens, attach to pathogens and enter the body, the cells are then targeted by killer T cells to be destroyed. However, cancer cells often appear as normal, healthy cells to the body and seem "innocent" like sheep. Due to this, they are not immediately recognized as dangerous by the body's immune system. Cancer cells have unique antigens called, Tumor-Specific Antigens (TSA), which are the wolves (NCI). Cancer cells will release programmed death-ligand 1 (PD-L1) to inactivate the killer T cells from recognizing and destroying the malignant cells (Ribas). Thus, cancer cells can avoid detection from the immune system. Without detection, cancer cells will begin to rapidly reproduce and could lead to metastasis. Cancer cells will replicate so quickly that their cells will not be fully developed (CancerResearchUK). These immature cells are increasingly likely to have more mistakes and will be a vicious cycle of producing more cancer cells, more quickly.

Why is CRISPR-Cas9 Better Than Cancer Treatments Now?
Cancer kills but so do the current treatments. Treatments like chemotherapy are employed to kill all fast-growing cells within the body but it cannot tell the difference between healthy and cancerous cells (ACS). This means that cancer cells and healthy cells will be damaged or destroyed. Consequently, cancer patients experience symptoms of hair loss, weakened immune systems, and fatigue (ACS). The advantage of the CRSIPR-Cas9 technology is that it can target a specifically mutated cell and hinder that cell from replicating. Thus, allowing the other healthy cells to continue to replicate and will not create burdenous symptoms. Another way that CRISPR-Cas9 can effectively treat cancer was shown in a study conducted in 2017 that explains that CRISPR-Cas9 can influence killer T cells and increase their recognizability of TSA (Baylis). Therefore, eliminating the opportunity for cancer cells to continue to replicate and allow the immune system to fight back. Overall, the specific nature of CRISPR-Cas9 proves to be an efficient tool when trying to fight cancer quickly. This could mean increased survival rates or longevity of life.
Moreover, it is important for cancer treatment symptoms to be reduced to improve the quality of life for cancer patients. Cancer patients already must come to terms with their diagnosis and endure difficult, invasive treatments. Therefore, minimizing the side-effects could dramatically improve their quality of life. By improving quality of life, it can improve chances of survival (Latifi).

Environmental Uses of CRISPR-Cas9
The tracrRNA discovery ignited the use of CRISPR-Cas9 technology to be applied to biomedical engineering on humans (Liao). Before tracrRNA was discovered, CRISPR-Cas9 was used to help fix ecological problems. One of the first uses that commercialized CRISPR was by Danisco in 2008 on a dairy farm (Cohen). CRISPR was used to alter S. thermophilus, to make the bacteria resistant to bacteriophages that otherwise would have caused the yogurt and cheese produced on the farm to spoil quickly. The genetic engineering resulted in the longevity of dairy products shelf-life (Cohen). This was optimal for farmers because they had fewer resources going to waste as well as gaining more money. Effectively, CRISPR was advantageous as it was able to generate more capital for a lower cost than the competing, more difficult to use technologies. However, environmentalists questioned if these practices were ethical and what effects they would have on human health (Singh).

Potential Adverse Effects of CRISPR-Cas9
Scientists hadn't begun to test CRISPR-Cas9 on human embryos until 2015 (Cyranoski). Listen to this podcast discuss the first human trial of CRISPR-Cas9 in the United States and to learn more information. It is important to note that the long-term effects of using genetic engineering on humans are unknown. Overall, CRISPR-Cas9 technology needs to be further researched in humans as it is relatively new to human application.
As heard in the podcast, there are instances where healthy cells can be genetically modified but create no side-effects. CRISPR-Cas9 technology has wonderful precision but lacks accuracy at times. This means that it delivers the alterations consistently, but it may target cells that are not meant to be targeted. Therefore, healthy DNA in cells may change in function and could cause lasting effects. The purpose is to fix mutations and to not create new ones. But to help treat cancer, is it worth the risk?
