Strong Placebo Effects in ED Drug Trials
Tumblr media
MedicalResearch.com Interview with: Alexander Stridh, MSc Department of Clinical Neuroscience Karolinska Institute Solna, Sweden MedicalResearch.com: What is the background for this study? What are the main findings? Response: We wanted to investigate how large the effect size is in the placebo arm of clinical trials with PDE5Is for erectile dysfunction. The major findings in this study is that there is a placebo response in general in ED, though it is hard to tell if this is a genuine placebo effect rather than normal fluctuations in symptom severity among patients. Another interesting finding was that the placebo response seems largely more important when the cause of ED is mainly due to psychogenic factors, as in post traumatic stress disorder. Lastly there was an important finding that there was no difference between placebo and active drug with daily treatment of PDE5Is  for long term recovery of erectile function after prostate cancer treatment, a practice which is common in some places and seems to be questionable.  MedicalResearch.com: What should readers take away from your report? Response: It is clear that Viagra and other PDE5Is work very well in many cases, but we should not disregard the importance of psychological aspects of ED. Particularly in individuals who have no clear physiological causes for ED. Some patients might benefit better from other interventions, e.g. psychotherapy, than the standard pharmaceutical interventions depending on the individual cause of ED.  MedicalResearch.com: What recommendations do you have for future research as a result of this work? Response: I would like to see more focus on the cause of the ED in the individual case and that research into treatments are based on this. All drugs have side effects, which in some cases can be severe, and should only be used when absolutely needed. I would also like to see that the practice of daily treatment with PDE5Is for recovery of erectile function is put into more question since it does not seem like this practice is backed up with solid evidence. It would be of great value if more clinical trials involved a third non-intervention arm that the placebo arm could be compared to. This would provide a better understanding of what would be a placebo effect and what are unspecific responses related to natural fluctuations in symptom severity of a disease. Earnings Update: Second GW Pharmaceuticals Stock Price Increase After February's Alzheimer's Trial News
Tumblr media
GW Pharmaceuticals (NASDAQ: GWPH) has been on a tear over the past month after exceeding expectations with its Q4 results, which were released at the end of February.
GW By the Numbers
Revenue increased 68% year on year to $6.8 million, comfortably ahead of Wall Street’s anticipated $5.4 million. Net loss of $0.20 per share was in line with its performance the previous year, and again it beat the consensus estimate. The operational highlight in Q4 was a strong launch of its flagship medicinal cannabis product, Epidiolex, in the US. It is the first CBD drug ever approved by the FDA for Dravet syndrome, a form of epilepsy, and more than 500 US doctors prescribed it in November and December 2018. That led to net sales of $4.7 million for the two-month launch period, and GW Pharmaceuticals anticipates soaring demand in the months ahead. It reported that filled prescription is up 150% in January 2019 compared to the previous month, with demand coming from major centres of excellence and local epilepsy clinics. In December, it met with the FDA about the potential for using Sativex to treat patients with Multiple Sclerosis spasticity, and it will begin a pivotal study in Q4 2019 on this. It is also undertaking a clinical trial Tuberous Sclerosis Complex, with data expected in Q2 2019. GW Pharma has a commercial footprint in place in five major European markets, and it expects the results of its European submission for Epidiolex by Q2 2019 (learn about another company capitalizing on European markets here).
The Future of Pharmaceuticals is Bright for GW
The firm is set to enjoy continued momentum in 2019 as there is an unmet medical need to treat Dravet syndrome. It is also diversifying by launching trials into treating different conditions, and investors are impressed. GW Pharmaceuticals Medicinal Cannabis Drug Trials

Source: GW Pharmaceuticals Earnings Presentation Tekla Capital Management LLC raised its holdings in GW by 45.3% during Q4, while Resources Investment Advisors Inc. and Horrell Capital Management Inc. both lifted their shares. NumerixS Investment Technologies Inc and Oppenheimer Asset Management Inc. both purchased new positions in the firm. The share price was $97.39 on December 31, 2019, and it increased to $152.89 by February 26. It then saw a sharp increase to $173.99 after its Q4 results were announced, and it has now settled at $166.57 this week. It has a strong balance sheet, with plenty of growth potential, so it is likely to remain a popular stock among investors in the year ahead.
Recall - Not the Only Recent Stock Price Spike For GW
GW Pharmaceuticals saw its share price increase in mid-February after Alzheimer’s Research UK announced a cannabis trial with the British firm. Cambridge-based GW Pharmaceuticals produces a cannabis mouth spray called Sativex that contains equal amounts of THC and CBD. It became the first cannabis-based medicine to be licensed in the UK and it is used to treat symptoms of multiple sclerosis, including spasticity, pain, tremors, and sleep disturbance. Now the respected charity Alzheimer’s Research UK has decided to fund a pioneering clinical trial at King’s College in London to discover the efficacy of Sativex in treating dementia sufferers. It said that around half of the 850,000 Brits living with Alzheimer's will experience agitation or aggression, making it one of the most challenging symptoms of the illness. It hopes that cannabis can negate this. It is devoting around £300,000 ($384,000) to the trial, which will see a study team recruit volunteers aged between 55 and 90 who suffer from dementia and experience symptoms of aggression or agitation. They will take Sativex for four weeks and, if their symptoms improve, they will perform a much larger clinical trial. Right now only MS patients can be prescribed Sativex, but Alzheimer’s Research UK will aim to change that if the spray proves to be effective. The researchers need to assess how practical it is to use a mouth spray if a patient is displaying extreme aggression. But chief scientific officer Dr. David Reynolds said no new dementia treatments have emerged in the past 15 years and that it is imperative for the medical community to test novel treatment methods in rigorous clinical trials. GW Pharmaceutical’s stock price increased to $1520.50 at the close of trading on February 15th. Cannabis is legal for medicinal purposes in the UK, but it has not taken off to the extent seen in countries like Germany. That is due to the strictness of guidelines that doctors must adhere to, but the landscape is starting to change and this trial could help unlock the industry. Read the full article

HEADCANON - CORTEXIPHAN TRIALS I.
Olivia Dunham had been subjected to trials of the non-FDA approved drug Cortexiphan created by Walter Bishop and William Bell. It started when she was 4 years old, and she had been signed up by her mother, much to the chagrin of her abusive step-father. Of course, there were reasons for this. Her mother, who died some time after, wanted to provide Olivia some sort of reprieve from the chaos of her home life, from the things which would happen behind that closed red door. It became very apparent very early on that young Olivia would be special, but not even the scientists would realize just how special she would be. It was not long after the trial started that SUBJECT 13, as they would call her, would be seen as an anomaly. The doses started small of course at first, easing these young minds into what was to come. Later that same year, she had crossed over for the first time --- crossed over into the other side. What she saw terrified her to such a degree that she set the room on fire. To this day, the walls still are scorched by her fearful flames. The trials would continue for years, and as time would go on, the young Olivia Dunham would prove to exhibit abilities far beyond the men’s imaginations. The others, sure, they would display profound abilities but none quite like her. She had immense power, power forged from extreme emotion and as she got older it would only continue to persist. As she would enter pubescence, the doses would increase. The more of the drug that would be introduced, the more vehement her ability and the more unpredictable she would become. It would be intense, volatile. The facility would succumb to several repairs for them to realize this. Her fear would transform into anger. Her sorrow would become guilt. It would become apparent that she could not continue on in her double life; so to speak. Her home life was not safe. Not just for herself, but for her step-father, who would eventually find himself dead ( to be elaborated later ). Her school life would not be able to continue, for her disruptive behavior and unspeakable incidents would follow her there, leading to several expulsions and suspensions ( to be elaborated later as well ). So the JACKSONVILLE FAMILY DAYCARE CENTER would become her entire existence, and she would consistently prove herself to be more than what Walter and William could have imagined her to be. Although it would not come without a price. Young Olivia would not know what it was like to have a normal childhood or adolescence, she would succumb to migraines in extremus after utilizing her abilities for extended periods of time. High doses of Cortexiphan would induce seizure activity. She would have extreme emotional dysregulation. She would have severe PTSD, just from the tests ( her childhood trauma experienced from her step-father a whole other can of worms ). She would have the gift / curse of extraordinary recall.  Trust would be exceedingly hard for our young Olivia to come by. And when trying valiantly to contain her power, she would become severely hot --- reaching temperatures unsustainable by any typical human. She would also succumb to fainting spells, and bouts of dizziness. However, she would not be affected by crossing into other universes as those not treated with Cortexiphan would.
MORE WILL BE REVEALED.

It was a small study with only 18 rectal cancer patients who took the same drug. However, the results were surprising. In the drug trial, cancer disappeared in all patients. It was undetectable by physical examination, endoscopy, PET scans, as well as MRIs. Luis A.      ❝ I always thought Ivankov’s devil fruit powers were interesting. ❞ Yes, indeed - the actual physical aspect of it. If Caesar could harvest that power, then he could add a whole new dimension to his drugs, and, of course, his ongoing drugtrial which would make giants would be an easy task if he could get his hands on growth hormones. Ah, well, this was all just on his wishlist. He doubted he’d ever even meet Ivankov, so, for now, Kuma would have to do.

SEND A WORD OR PHRASE ANONYMOUSLY OR NOT AND I WILL WRITE A HEAD CANON SURROUNDING IT FOR MY MUSE.
@anonymous sent: DRUGS.
From the age of 4 years old, Olivia Dunham has been subjected to drugs. Of course, this is not by means of recreational or addictive use --- no, rather something different, and perchance even more heinous. It was one thing to be exposed to the life of addiction, of course, a lifestyle that is truly disparaging for any child to have to endure. However, to be willingly submitted by one’s parents to an experimental trial for a drug that was not by any means FDA approved yet, well, that was a cruel, cruel fate. She was subjected to things that no child, let alone a child THAT SMALL should ever have to experience.
Cortexiphan became a part of her long before she could have any intellectual or emotional understanding of it. All she knew then was that these funny men in lab coats were giving her “MEDICINE” and made peculiar expressions to try to give levity to a situation that was rather dismal. As she got older, the doses increased, and she would come to understand just what was going on. She would have all sorts of wires, tubes hooked to her, IV’s inserted on a nearly daily ( if not indeed daily ) basis. For years, her arms and hands were riddled with poke marks, not too much unlike that of an IV drug user.
But, outside of that, Olivia Dunham was not one to take illicit substances. Although, one could argue that she would have every reason to do so if they knew exactly what she had been through. As a teen, she would on the very rare occasion smoke weed ( no more than a couple times a year ), but aside from this the only thing she does on a regular basis is alcohol. But not to the point of excess, or in any addictive capacity. And then as an adult, she would be administered drugs in order to aid in Walter’s scientific findings for a case, or if she had to go into the water tank ( situations akin to but not limited to this ).
