The brotherly moments are what I’m looking forward to the most for the sequel. I’m hoping we can potentially get a scene that will top the reunion scene. Are there certain kinds of brotherly love moments you’d like to see in the sequel?
Big same. What I'm most looking forward to is all the little subtle shows of affection: headlocks, side hugs, cap tugs, belly pokes, shoulder nudges, jabs, jostles... all the bits of body language that made their relationship feel so close and real in the first movie. And of course, the return of that silly little secret handshake.

Another thing I want to see is more of Mario's protectiveness. I know some are hoping that he gets a little overprotective after being separated from his bro in the first movie, but if we don't get that I want to at least see Mario's pre-adventure protectiveness.

Or the kind of protectiveness he exhibited in the Super Mario Adventures comics, where even though he's focused on the fight he always keeps an eye on his brother, and the moment Luigi calls out for help he immediately becomes first priority.

On the flip side, I want to see more of Luigi being the emotional support/voice of reason. He may be a ball of nerves in moments of peril, but he's proven to be very levelheaded overall. Since I picture the arc of the sequel being Mario slowly coming to embrace the role of a true hero– with all the responsibilities that come with it– it'd be nice if he had a low moment of feeling like a failure that Luigi talks him out of.

Now, if they're going to try and top the reunion scene in the first movie, there is one surefire way to do that: slow down. As good as that hug was, there was so much going on that it was hard to feel the full impact on the first watch. Quiet things down for a moment... pump the breaks... focus in on the facial expressions... let us really feel the hug. Throw in another forehead touch while you're at it.
But you know what I'm really really excited to see? Sword and shield fight dynamic. I desperately want to see them fighting side-by-side without the superstar! Give me Mario spearheading the charge while Luigi watches his blindside! Mario throwing himself mindlessly into a dangerous stunt so long as Luigi is there to catch him, Mario fighting in close combat while Luigi draws the target's attention with long-range attacks, both of them falling into perfect synch when they need to deliver an extra-powerful blow.
I am so aggressively obsessed with the idea of their character traits being fully exhibited and explored through the way they approach a fight, you have no idea.

ik it’s probably a classic case of extrapolating way too much but... the way raph’s character design lines up w his whole arc and role and struggles...
like the fact that he is so so so much bigger than the others. raph is big, raph is strong, raph is steady and sturdy and he can literally pick up his entire family and carry them all at once. 
and like, when raph is so big and so strong and such a reliable thing. when raph is the protector, the one calling the shots on missions, the mother hen, the first point of authority. when raph is there, overprotective, when raph (for all that his brothers poke at him not being good under pressure) always always ALWAYS comes through at the end of the day when things are serious, ALWAYS gives it everything he’s got. 
his design and his learned role/behaviors in this family are just the perfect storm of why it took up to the season finale to drive home the issue.
so much of the series carries the default energy of “raph will handle it.”
raph will hold up the ceiling above you. raph will throw himself over you and take a hit and get back up and keep fighting. raph has a power that makes him even bigger and draws more attention and makes him able to carry MORE. raph will be the substitute parent. raph will be put into the mentor role through leo’s leadership arc. 
and raph is big. he’s built to carry heavy loads. raph is strong. raph is bold and loud and always ready to try to push on. even if he doesn’t know what to do or what he’s doing, he won’t give up and we’ll all pull together and things will turn out okay.
(his room is full of teddy bears. he dipped out on a mission to try to take a picture of a pigeon carrying a slice of pizza. he’s terrified of being alone.
he’s just as much of a kid as his brothers are. he’s just as new and inexperienced with the things happening to them as his brothers are. but for him, for some reason, there’s like this double standard where that becomes a huge glaring flaw.)
idk this got very sloppy and uncoordinated. i’m very in my feelings about raph right now though.

I love the secret after-credits scene of the Mario movie where Bowser tells Mario that he figured out that even if he had Peach, he wouldn't be happy. He figured it out that love is trust on both sides of a relationship (and that real love isn't just a one-sided thing) and he figured out that Mario's brother that he affectionately calls "Greenie" actually makes him happy because Luigi found it in his heart to trust the big koopa. Bowser also said he was sorry for wrecking Brooklyn and he's actually taking anger management classes now (with extra support from Luigi).

So @kuramirocket got me thinking of a Vampire AU for the Mario Bros (while discussing in this post) so I figured I would share some thoughts of how I personally would want it to go:
please note that I am not really a big vampire enthusiast, nor do I know that much about vampire lore. I never got into Twilight, nor have I read Dracula (yet). So I'll only be referring to the basic stuff I know about vampires. For those interested: do with this au as you please :)
- Luigi gets turned into a vampire (whether by dark magic, an item, or another vampire). And it's not long after that effects start to take place (his skin burning in the sun, a taste for blood, the urgent need to wander about in the night). His skin also gets a more pale tinge and his eye colour turns purple.
- Luig tries to keep this hidden as long as possible, but Mario finds out and is devasted. He blames himself for not being able to protect his brother from this. He promises to fix this and help Luigi find a cure.
- After many months of travelling (and trying to keep Luigi's new vampire instincts in check in the meantime), they soon come to learn the truth that there is no cure. Luigi is stuck like this forever.
- Mario, without a shred of doubt, asks Luigi to bite him, so he can become a vampire and live with him in immortality.
- Luigi is of course furiously against this. Why should Mario suffer because of him? His brother deserves much better than this, and he should just forget him.
- Mario gets upset at this and firmly tells him that he wants this. Luigi has done nothing wrong, and if fate is deciding that there is no way out of this, Mario would rather shoulder that with him. They're a team, always together, and he can't bare the idea of one having to watch the other age...
- Luigi finally agrees. They hold each other tight, and Luigi (after some hesitation) bites down on Mario's neck.
- It's so painful. And Mario can feel the venom swarming around inside his body. But he knows it's not Luigi's fault. He holds him closer, trying to communicate via his touch that this is fine, everything is ok, this is what he wants.
- After some time Luigi stops and Mario steps back. He's lightheaded, his skin already turning pale, and his eyes becoming a shade of purple. He also has two bite marks on his neck (which he forever carries around as scars). But he's ok. Mario smiles at Luigi reassuringly, knowing it worked.
- They move far away and live in immortality together, deciding to become dark shadow hunters (to protect innocents) and only live on a diet of animal blood and plant sap.
